　　来源于水母Aequoreavictoria的绿色荧光蛋白 ( greenfluorescentprotein ,GFP)现已成为在生物化学和细胞生物学中研究和开发应用得最广泛的蛋白质之一 .其内源荧光基团在受到紫外光或蓝光激发时 (λmax=395nm ,小峰在 4 79nm )可高效发射清晰可见的绿光 .GFP的高分辨率晶体结构为了解和研究蛋白质结构和光谱学功能关系提供了一个极好的机会 .GFP已成为一个监测在完整细胞和组织内基因表达和蛋白质定位的理想标记 .通过突变和蛋白质工程构建的GFP嵌合蛋白在生理指示剂、生物传感器、光化学领域以及生产发光纤维等方面展示了广阔前景

　　绿色荧光蛋白(ｇｒｅｅｎｆｌｕｏｒｅｓｃｅｎｔｐｒｏｔｅｉｎ,ＧＦＰ)是一类存在于包括水母、水螅和珊瑚等腔肠动物体内的生物发光蛋白.当受到紫外或蓝光激发时,ＧＦＰ发射绿色荧光.其独特之处在于:它产生荧光无需底物或辅因子,发色团是其蛋白质一级序列固有的[1].来源于水母(Ａｅｑｕｏｒｅａｖｉｃｔｏｒｉａ)的ＧＦＰ由238个氨基酸残基组成,分子质量为27ｋｕ[2].水母的ＧＦＰ基因是目前唯一已被克隆的ＧＦＰ基因[3].本文主要综述关于这种ＧＦＰ的研究.　　水母ＧＦＰ的荧光很稳定,无种属限制,已在网柄菌、大肠杆菌、酵母及多种动植物…

Green Fluorescent Protein.

　Recently the green fluorescent protein (GFP) from the jellyfish Aequorea victoria has emerged . It has been widely studied and used in biochemistry and cell biology. The protein emits green light ( λ max =508 nm) when excited with ultraviolet (UV) or blue light ( λ max =395 nm, minor peak at 479 nm). It is extremely useful as a marker for monitoring gene expression and as a tag in studying protein localization in organisms, intact cells and organelle. Mutagenesis and engineering of GFP fused into chimeric proteins are opening new prospect in physiological indicators, biosensors, photo chemistry and production of luminescent fiber.
